Interstitial
deletion of chromosome 15 is
responsible for approximately 70-75%
of cases of PWS. [Robinson 1991,
Smith 2003]

The
deletion which includes around 4 Mb
of DNA extends from D15S9 through to
D15S12. The exact mechanism of the
deletion is not fully understood
although research has located
different breakage sites along the
chromosome. One study confirmed the
presence of two proximal breakage
sites on the chromosome, one lying
between D15S41/S542 and D15S543 and
the other being proximal to
D15S41/S542.
